Effect of N-methyl-N-nitrosourea on the protein-synthesizing system in mouse liver and hepatoma 22a cells.

چکیده
SUMMARY A single injection of nitrosomethylurea (80 mg/kg body weight) to mice bearing hepatoma 22a led to an injury of the main components of the protein-synthesizing system (poly ribosomal complex and soluble factors of the cell sap) in liver and hepatoma cells. In liver cells of adult animals, the activity of the protein-synthesizing system returns to a normal level 5 to 15 hr after injection, while in hepatoma the process is irreversi ble for at least 48 hr. These results were confirmed and extended in experiments conducted in vitro with cell-free protein-synthesizing systems.

Mismatch repair and differential sensitivity of mouse and human cells to methylating agents.
The long-patch mismatch repair pathway contributes to the cytotoxic effect of methylating agents and loss of this pathway confers tolerance to DNA methylation damage. Two methylation-tolerant mouse cell lines were identified and were shown to be defective in the MSH2 protein by in vitro mismatch repair assay. A normal copy of the human MSH2 gene, introduced by transfer of human chromosome 2, re...
متن کاملInhibition of protein synthesis by N-methyl-N-nitrosourea in vivo.
1. The intraperitoneal injection of N-methyl-N-nitrosourea (100mg/kg) caused a partial inhibition of protein synthesis in several organs of the rat, the maximum effect occurring after 2-3h. 2. In the liver the inhibition of protein synthesis was paralleled by a marked disaggregation of polyribosomes and an increase in ribosome monomers and ribosomal subunits. No significant breakdown of polyrib...

متن کاملGene Expression under F8 Promoter Driving In Mouse Hepatoma Cells: A Step towards Gene Therapy of Hemophilia
Background and Objectives: Significant progress has been made in treatment of hemophilia. Ex-vivo gene therapy is going popular due to the capability of this method in using isogenic cells for genetic manipulation and reintroducing them into same host after proliferation. Most gene therapy techniques use viral vectors, which usually harbor a strong and non-specific promoter (e...
